I’ve started running Instagram ads, but when someone comments on the actual ad rather than the original post in my feed, the ManyChat automation I set up doesn’t work. Can anyone help with this?

That's a limitation between Meta and Manychat.
The recommended way for this case would be creating some exclusive creatives for your ads and use the Ad Trigger:
