I lose a lot of leads on Instagram, because their message is not being delivered to their DMs - they just see a blank message.
To make this situation better, I want to send a follow up DM if they didn’t click on the ‘comment reply message’s button after 5-10 minutes.
How can I do that is this even possible? Thanks.

Unfortunately, that isn't possible due to Meta's restrictions with this kind of automation. The contact must click on a button/smart reply to initiate a conversation window. Only after that it's possible to do some kind of follow up.
I'd suggest you tr to rebuild your first message with buttons and/or quick replies and check if anything runs better. You can even randomize the first message to create kind of a A/B Test. 😉
