I was just told the Comment Reply can work on Meta Ads Manager if you choose an existing post to run as an ad. Is that true?

Yes, but only in this scenario.
I also had some cases in wich it didn't work (Meta issues)… Bu it should work as expected.

It looks like Meta handles Ads and Posts like two totally different things. Because of that, Manychat can't receive any information about comments on Ads.
The alternative is using the Instagram/Facebook Ads Trigger = https://help.manychat.com/hc/en-us/articles/14281321942428-Instagram-Ads-Trigger
