It also works with the Free version, but with a lot of limitations (Check here: https://manychat.com/pricing).

Once you're in the Trial Period, almost all PRO features will work and you will be able to build more advanced automations than in the Free version.

As you're starting with Manychat, I suggest starting with some templates Manychat offers. In the image above I've marked both templates that you can use to respond and DM users that comment in your Reel.

There are two types of automations:

• Quick Automation: Is the easiest to implement. Is like filling out some form with a welcome message and buttons with links to deliver some content. As much as it simplifies your life, it also limits a lot of possibilities. 
• Flow Builder: Here you access all of Manychat's power and are able to build complex and resourceful automations that interact with your contacts, collect data, check conditions and so on. The template is very straightforward and easy to understand.

If you're just willing to put an automation to run and have the least work as possible, then go with the Quick Automation, but if you're willing to learn something about automation, go with the Flow Builder… the learning curve is a little steeper, but you will thank me later.

Yes, you can do it.
On the trigger you have to choose the option “next post or reel” and then publish your reel and the automation will work
